The US Food and Drug Administration has approved ozanimod for adults with moderately to severely active ulcerative colitis , the company has announced.

Ozanimod , an oral medication taken once daily, is the first sphingosine-1-phosphate receptor modulator approved for UC.

In March 2020, the FDA approved ozanimod for adults with relapsing forms of multiple sclerosis, as reported by Medscape Medical News.

The approval of ozanimod for UC is based on data from True North, a phase 3 placebo-controlled trial that evaluated the drug as induction therapy over 10 weeks in 645 adults with moderately to severely active UC, followed by maintenance therapy over 42 weeks.

All participants in the trial had failed to respond adequately to or were intolerant of oral aminosalicylates, corticosteroids, immunomodulators, or a biologic 30% had previously experienced treatment failure or were intolerant of tumor necrosis factor blockers. Prior to and during induction, patients were treated with oral aminosalicylates and/or corticosteroids.

During induction at week 10, the trial met its primary endpoint significantly more patients who took ozanimod achieved clinical remission than patients who took placebo .

Ozanimod was also superior to placebo on the secondary endpoints of clinical response , endoscopic improvement , and endoscopic-histologic mucosal improvement .

In vivo MRI of mice was conducted using an 11.7 T vertical bore scanner . Anesthesia was initially induced with 2.0% isoflurane and maintained with 1.6% isoflurane during MRI. The body temperature of the mice was maintained at 37 °C with circulating warm water. T2 weighted axial and sagittal images with fat suppression were obtained using the rapid acquisition with relaxation enhancement technique. The acquisition parameters were as follows: field of view =25 mm×25 mm, matrix size=256×256, in-plane resolution=98 m, slice thickness=500 m for axial images and 300 m for sagittal images, repetition time =6000 ms, echo time =36.7 ms, number of averages =8, and acquisition time =13 min. MRA was performed using the 2D time-of-flight technique. The acquisition parameters were as follows: FOV=25 mm×25 mm, matrix size=256×256, in-plane resolution=98 m, slice thickness=400 m, slice pitch=250 m, TR=20 ms, TE=2.7 ms, NA=4, and TA=16 min. The volume rendering method was performed with the acquired images to visualize blood vessels three dimensionally. High signal intensity regions in the obtained images indicated the inflammation of colon. The pixels of inflamed edematous regions with high signal intensity in the axial image were measured using the Image J 1.52a software , and a comparative analysis between the control group and Si-based agent-treated group was conducted.
